This thesis focused on unraveling metabolic reprogramming in human tumours. In particular, the study presented focuses on PPARγ signalling network in BRAFV600E driven colorectal cancers through regulation of peroxisomal phospholipid biosynthesis pathways. PPARγ mediated regulation of peroxisomal lipid metabolism was shown as a key regulator of intracellular lipid storage and further drove chemoresistance and cancer stem like phenotypes. Altered tumour metabolism is an important mechanism of drug resistant tumours and targeting tumour metabolism provides and exciting therapeutic opportunity.
