This thesis describes the development of approaches to study reservoirs of silenced HIV in an important but understudied cell type, macrophages. This includes the development of methods which may be used for clinical applications to test the effects of potential therapeutics. The use of these approaches also advances understanding of the processes underlying the silencing of HIV reservoirs. Silent HIV reservoirs are the greatest barrier to HIV cure. The work presented in this thesis addresses these reservoirs in macrophages and thus informs cure strategies which target HIV reservoirs.
