Synergistic killing mechanisms of novel polymyxin B combination therapies against Pseudomonas aeruginosa

The global health system is facing a big threat due to dissemination of highly resistant bacteria namely Superbugs . This is accompanied with a dry up of antibiotics industry pipeline. Polymyxins are only treatment option available for treatment of these Superbugs; however, these notorious pathogens find ways to resist last line therapy polymyxins. Combining of polymyxins with nonantibiotics compounds has emerged as cost-effective way to save the efficacy of last line polymyxins and reduce resistance. We identified effective novel polymyxin combinations to fight superbugs by using systems pharmacology approach. These novel combinations preserve the clinical efficacy of last resort polymyxins and prevent polymyxin resistance.
