Transient Receptor Potential Vanilloid 4 (TRPV4) is a calcium-permeable ion channel that contributes to many important physiological processes in the body. Despite intensive investigation, a precise mechanistic understanding of how TRPV4 contributes to health and disease is lacking. To address this, sensitive mass spectrometry and analytical approaches were employed to reveal how TRPV4 function influences the proteome, metabolome and lipidome of macrophage cells within the innate immune system. This work revealed unappreciated roles for TRPV4 in macrophage homeostasis and the immune response. It significantly advances our understanding of TRPV4 function, to support new lines of investigation and drug discovery.
