Mechanisms of Renal Injury and Autoimmunity in ANCA Vasculitis

ANCA vasculitis is an autoimmune disease that commonly affects the kidney. This thesis examines two aspects of disease pathogenesis. One is whether the NLRP3 inflammasome, a recently identified part of the innate immune system, contributes to kidney disease in ANCA vasculitis. Experiments contained in this thesis show that blocking the NLRP3 inflammasome is protective in mouse models of disease, and the mechanisms behind the protection seen are explored. The second part of the thesis focusses on the use of monoclonal antibodies to make improved mouse models of ANCA vasculitis.
