Implementation and Correlative Application of Advanced Microscopies for Imaging Cellular Ultrastructure

Super-resolution fluorescence imaging allows for visualisation of cellular structures and sub-cellular organelles with resolution much improved over the diffraction limit of light. While extensive detail concerning the target in question can be obtained using these methods, the wider context is not visible. Correlative application with other techniques such as atomic force microscopy can place a structure of interest within its surrounding environment, yielding greater insights. This thesis develops the correlation of live and fixed cell super-resolution fluorescence imaging with atomic force microscopy for viewing intracellular structures, and describes the implementation of a bespoke lightsheet microscope for fast 3D cellular imaging.
