Triple-negative breast cancer (TNBC) is the most challenging breast cancer subtype to treat, and there is no viable targeted treatment strategy. Conventional TNBC chemotherapy is limited by its nonspecific damage to normal tissues and organs. This thesis describes the development of a novel porous silicon drug-delivery nanotherapeutics, which is TNBC-specific and biodegradable. The result indicates an enhanced tumor accumulation of nanoformulation inducing selective TNBC killing without much damage to normal tissues. The potential subcellular mechanisms of the TNBC targeting are clarified to understand the targeting mechanism. This study builds a compelling case for taking TNBC therapy from bench to bedside.
