Development and Evaluation of Inorganic-organic Hybrid Nanoparticles for Facilitating Delivery of mRNAs and siRNAs Into Breast Cancer Cells

Breast cancer at an advanced stage cannot be curable with available treatment. In this regard, RNA therapeutics emerged as an alternative therapeutic option for managing cancer, even at an advanced stage. RNA therapeutics in the form of messenger RNA (mRNA) or small interfering RNA (siRNA) are employed to restore or silence the cardinal gene responsible for the development and metastasis of cancers. Systemic circulation, delivery of naked mRNA or siRNA is not advantageous since nucleases can degrade the free nucleic acid. Considering the safety and efficacy issues, none of the existing vectors for the delivery of mRNA or siRNA is ideal for clinical use. Thus, we developed a SSNs –LNPs-based hybrid nanocarrier for combined delivery of mRNA and siRNA to improve their therapeutic efficacy against breast cancer cells both in vitro and in vivo.
