Fluorescence microscopy has a resolution limit due to the diffraction of light, which hinders accurate visualisation of nanoscopic organelles and features within the cell. Super-resolution microscopies overcome this limitation using innovative strategies, including single molecule localisations, correlating fluorescence fluctuations, and physical expansion. Each provides a unique advantage for subcellular biology research, such as achieving resolutions as good as 20 nm, the ability to capture live cell dynamics, and decrowd the dense cellular environment. This thesis presents the development of super-resolution microscopy methods dSTORM, SOFI and ExM, and their applications in studying viral protein mechanisms, anticancer drug effects, and genomic organization.
