This thesis explores two drug hypersensitivity reactions linked with the risk gene variant HLA-B*57:01: pazopanib-induced liver injury and abacavir hypersensitivity syndrome. It establishes, for the first time, a mechanistic role for HLA-B*57:01 in pazopanib-induced hypersensitivity, and describes two abacavir-induced T cell epitopes. This work suggests distinct modes of drug-HLA-B*57:01 interaction underlie these unwanted immune reactions.
